中文摘要: 肿瘤转移为乳腺癌患者死亡的重要原因,而循环肿瘤细胞(CTCs)的检测能够为潜在转移提供早期诊断。与其他CTC富集装置相比较,基于纳米结构材料的CTC捕获/鉴定装置能够呈现一种生物学中纳米尺度相互作用的独特概念。基于我们的前期工作,运用包覆细胞捕获(anti-EpCAM)的3D纳米纤维多孔支架作为新型CTC捕获/鉴定装置。本课题的主要内容是:1)构建CTC捕获/鉴定装置 2)评估CTC捕获/鉴定装置细胞捕获效率 3)鉴定CTC捕获/鉴定装置所捕获的CTCs并研究该装置临床应用性。本课题研究意义为:通过构建用于捕获和鉴定乳腺癌患者周边血液中循环肿瘤细胞的新型CTC捕获/鉴定装置,为乳腺癌患者提供更高效早期诊断的方法。

英文摘要: Given the fact that the most important cause of cancer-related death in breast cancer patients is metastasis, detection of circulating tumor cells(CTCs)could provide earlier diagnosis to potentially fatal metastases. In contrast to the other CTC capture approaches, nanostructured materials-based CTC capture/detection approach represents a unique concept that mimics the nature of nanoscale.interactions observed ubiquitously in biology. We introduce a new CTC capture/detection assay based on 3D nanofiber scaffold,which was grafted with cell capture agent (i.e., anti-EpCAM). The following specific aims will be pursued: 1) to fabricate the CTC capture/detection assay,2)to test the capture efficiency of CTC capture/detection assay,and 3)to detect the CTCs captured through the CTC capture/detection assay and to determine the potential application in clinic. The significance of this project is to provide a more efficient method for early diagnosis through fabricating a new CTC capture/detection assay for capturing and detecting CTCs from peripheral blood samples of breast cancer patients.
