中文摘要: 昼夜节律是指生物体的生理、生化和行为以24小时为周期的振荡。它是生物界最普遍存在的一种节律。正是由于生物钟的存在,使得机体在昼夜 24 小时中不同时相对同一药物、毒物的敏感性不同。氰戊菊酯(Fen)是一种重要的环境化学污染物,影响睾酮合成是其生殖毒性的主要表现之一。研究表明Fen对睾酮的影响具有昼夜时间差异,这种时间毒性可能与钟基因Bmal1相关,但具体的调控机制仍不十分明了。本研究拟以Fen对睾酮影响的昼夜节律差异为切入点,以Ca2+ -CaKM/RORα通路和miR-494-3p为轴线,利用整体动物和体外细胞实验模型,应用RNAi、报告基因及转基因技术研究氰戊菊酯对睾酮的昼夜毒性中钟基因Bmal1的作用,以及RORα、miRNA在此过程中的调控机制。本研究将为阐明氰戊菊酯的时间毒性机制提供重要参考、为合理使用氰戊菊酯提供重要的理论依据。

英文摘要: Circadian rhythms are the approximate 24-hour oscillations in behavioural or physiological processes that allow organisms to anticipate routine environmental changes and prepare for the appropriate alignment in order to adapt.fenvalerate is an important environmental chemical pollutants, and has the adverse effects on testosterone. We found that animals exposured during the daytime were more sensitive to fenvalerate than animals exposured at night, and Bmal1 may be involved in this process. However, the mechanisms of circadian sensitivity to the fenvalerate is still unclear. In this study, the methods of RNAi, report gene and transgenic technology are used to clarify the effect of Bmal1 on the circadian alterations of testosterone in male mice exposure to fenvalerate, and investigate whether the Ca2+ -CaKM/RORαpathway and/or miR-494-3p regulate this process. The project may provide the theoretical bases for rational using of fenvalerate.
