中文摘要: 随着生活、工作方式的改变及社会人口的老龄化,骨关节退行性病变已成为高发疾病,其中代表性疾病之一为骨性关节炎。骨性关节炎的发生与关节软骨损伤的病理密切相关。如何高效率地调控与恢复软骨基质蛋白代谢成为预防及治疗骨性关节炎的研究焦点。最新研究表明:骨性关节炎关节软骨和滑膜组织中异常增高的C/EBPB诱导MMPs的分泌,促进关节软骨的变性及破坏。由此,我们提出新的假说:调控C/EBPB基因表达及蛋白水平是靶向调节关节软骨细胞外基质代谢的一种有效途径,通过调节C/EBPB基因表达,抑制C/EBPB的蛋白水平和活性,即可达到抑制软骨退变的目的。本项目使用临床组织标本、细胞及动物模型,以分子生物学手法探讨C/EBPB在骨关节软骨基质合成和分泌中的分子调控机理,明确其在骨关节退行性病变中的作用,为骨关节退行性病变的生物学治疗提供新的途径及理论实验依据。

英文摘要: Osteoarthritis (OA) is a degenerative disease characterized by destruction of articular cartilage. However, no effective medical therapy to prevent OA cartilage destruction is currently available, owing to the limited understanding of the underlying molecular pathogenic mechanisms. The chondrocyte is a unique cell type in articular cartilage that is responsible for cartilage homeostasis and OA pathogenesis. Recently, It has been demonstrated that the C/EBPB is a catabolic regulator of cartilage destruction that acts by regulating the expression of various catabolic factors, including matrix-degrading enzymes. The regulation of C/EBPB gene and protein level in articular cartilage remains unknown. In this study, we attempted to explore the mechanisms in regulating the transcription and posttranscription of C/EBPB by using animal model of osteoarthritis and human chondrocytes, and confirm the roles of C/EBPB in joint degenerative disease.
