中文摘要: 多细胞生物各组织和器官的细胞数量取决于细胞的增殖和死亡,特定组织器官的细胞类型和细胞数量是相对恒定的。细胞生与死平衡的打破,是人类包括肿瘤在内的许多重大疾病发生、发展的基础。Salvador (人类的同源蛋白hSav1又称hWW45)同时参与了细胞增殖与细胞凋亡的调节,后续的研究将这一信号通路称为SWH信号通路,hSav1是其核心成员之一。我们的研究明确了蛋白HAX1和hSav1在正常结肠粘膜组织和肿瘤组织中的表达,结果表明蛋白hSav1和HAX1的表达水平与肿瘤的发生发展和预后密切相关。从而建立了蛋白hSav1及HAX1和肿瘤之间的联系,为进一步的研究提供了依据。完善了hSav1对细胞凋亡和增殖的双重调控作用及机制的研究,即hSav1通过与Hax1, Rb之间相互作用双重调控细胞凋亡和增殖。

英文摘要: The dysregulation of cell proliferation and apoptosis causes many human diseases, including tumor.SHW signaling pathway was reported recently to regulate both cell proliferation and apoptosis and play a critical role in the tumorigensis. hSav1 is one of the core components of SWH signaling pathway. Our previous data suggested hSav1 is involved in mitochdrial apoptotic pathway and may affect cell proliferation through association with a core component of cell cycle, RB. It gives us a clue as to how hSav1 regulates both cell proliferation and cell apoptosis.This project is going to assess mRNA and protein levels of hSav1 in clinical samples of cancer; And then to explore the function and of hSav1 in cell proliferation and apoptosis and its related mechnisms.Taken together, these findings establish an important role for hSav1 in regulation of both cell proliferation and apoptosis. Furthermore, our work may yield novel therapeutic target for the treatment of cancer.
