中文摘要: 干旱是限制作物生长发育和产量提高的主要逆境因素之一。抗旱相关NAC转录因子在植物干旱应答中起着重要的调控作用。本项目以干旱胁迫处理的玉米幼苗为材料,利用基于新一代高通量测序技术的RNA-Seq (Quantification) 测序结合生物信息学方法、Northern Blotting和Real-time PCR技术,筛选玉米全基因组抗旱相关NAC基因并解析其在转录水平的表达谱。同时结合干旱胁迫下抗旱玉米自交系中NAC基因的动态表达规律及生理生化指标、NAC基因遗传转化玉米,探讨NAC转录因子与玉米抗旱性的关系。本项目的开展将有助于揭示NAC转录调控植物干旱胁迫应答的分子机理,为更有效地寻找改良作物抗逆性的抗旱转录因子提供新的思路和实验依据。

英文摘要: Drought is one of the major environmental stresses that affect plant growth and corp yield. Drought responsive NAC transcription factors play important roles in modulating plant responses to drought stress. To investigate the drought responsive NAC gene family in maize genome, and resolve transcriptional profiles in maize seedlings under drought stress, RNA-Seq quantification approach based on next-generation Illumina sequencing technology, together with bioinformatical analysis, Northern Blotting and Real-time quantitative PCR analysis, will be used in the present project. And then, the relationship between NAC genes and drought resistance will be revealed in drought-tolerant maize lines, according to dynamic gene expression pattern and physiological and biochemical indexes. The project will aid to unravel the molecular mechanism of NAC genes drought response, and provide a new approach for finding transcription factors responsive to drought stress improving crop stress resistance.
