中文摘要: 札幌病毒(Sapovirus,SaV) 属杯状病毒科单股正链RNA病毒,是导致人、猪和水貂等多种易感动物罹患急性胃肠炎的重要非细菌性病原之一,主要经粪-口途径传播,对人畜健康构成一定威胁。研究表明,某些猪源SaV和人SaV毒株高度同源,这提示SaV可能为人畜共患病病原。迄今为止,SaV在宿主细胞表面的受体依然未知。有鉴于此,本课题以目前唯一能体外培养的猪SaV Cowden毒株为研究材料,测定经放射标记的SaV毒株与易感细胞LLC-PK在经酶及多种试剂处理前后的结合活性、感染性的变化以确定受体性质;VOPBA法确定受体分子量;免疫共沉淀、LC-MS等方法鉴定受体;病毒感染阻断实验和受体重建实验验证受体;分析受体在猪体内的组织分布和表达丰度。为揭示SaV的宿主特异性和组织亲嗜性,从分子水平阐明SaV病毒感染与免疫机理,探索SaV可能的跨种间感染与传播等科学问题提供理论依据。

英文摘要: Sapovirus(SaV ), a positive single strand RNA virus belonging to Caliciviridae,is one of the important non-bacterial pathogens cause human, pig and mink and other susceptible animals suffering from acute gastroenteritis. Fecal-oral transmission of the infection poses threats to human and animal health. Previous studies confirmed SaV strains isolated from porcine and humans are highly homologous, suggesting that SaV may be a zoonotic pathogen.To date, receptors for SaV infection on host cell surface the is still unknown. In view of this, this proposed study aims to use radiolabeled porcine SaV Cowden strain to infect LLC-PK cell. Receptor is initially investigated by enzymic digestion of the cells. VOPBA, coimmunoprecipitation combining with LC-MS are used to determine the receptor; Infection blocking experiment is used to verify the receptor; Recetpor distribution in vivo will be studied in order to reveal the SaV host specificity and tissue tropism.This study is hoped to explain the SaV virus infection and immune mechanism.
