中文摘要: 糖尿病是继心血管疾病和肿瘤之后的另一种严重危害人类健康的重要慢性疾病。目前全世界约有2.8亿糖尿病患者,其中90%是2型糖尿病;我国是糖尿病第一大国。对2型糖尿病发病机理研究、药物筛选和评价多是在啮齿类动物上进行的,由于种属关系,啮齿类2型糖尿病动物模型很难真实反映人类2型糖尿病。因此,寻找和创制低等的非人灵长类2型糖尿病动物模型备受关注和欢迎。树鼩是灵长类近亲或低等灵长类动物,本项目旨在开展树鼩基础生理代谢研究,获得作为疾病动物的基础生理数据。在此基础上,整合食物诱导、链脲佐菌素(streptozotocin,STZ)诱导、食物加STZ诱导等三种方法创建2型糖尿病树鼩模型。通过疾病同源性、行为表象一致性、药物治疗预见性等"三性"验证创制的2型糖尿病树鼩模型。为2型糖尿病发病机理的研究、糖尿病药物的研发提供新型低等灵长类动物模型。

英文摘要: Type 2 diabetes mellitus (T2D) is the third chronic disease killer after cardiovascular diseases and cancers. International Diabetes Federation (IDF) estimated 285 million people with diabetes worldwide, in which 90% were T2D in 2010. Additionally, Yang et al reported that China is the top one nation of T2D. Rodents have been widely used to explore the mechanisms of T2D as well as to screen and evaluate therapies of T2D. However, due to the far phylogenetic relationship, the rodent models of T2D only partially mimic the pathogenesis of human T2D. Therefore, new animal models of T2D, especially from lower non-human primates (NHP) or primate relatives, are urgently needed for T2D research. Tree shrew has been considered as a lower NHP or a close relative of primate, and have been widely used for biomedical research. We proposed this project to understand the basic biochemical and physiological characters of tree shrew, and then to focus on generation T2D of tree shrew via diets, streptozotocin (STZ), diets plus STZ. We will further verify T2D of tree shrew by reliability/replicability, validity, and generalizability. Our work will probably provide a new animal model of T2D for mechanisms exploration of T2D and evaluation of therapies of T2D.
