中文摘要: 为了研究民猪的抗寒特性,本项目将12头同期民仔猪随机分成2组,1组置于舍外饲养(-12℃#65289;,1组置于正常舍内饲养(15℃#65289;,处理24h后,收集血清,采用ELISA方法测定血清中胰岛素、肾上腺素、T3和T4的浓度;采集部分肌肉组织,利用Affymetrix的表达芯片,检测冷应激后差异表达基因。结果表明,冷应激24h后,实验组个体的血清中胰岛素含量出现明显的上升趋势,肾上腺素、T3和T4也出现上升趋势,但是与对照组相比,均未达到差异显著。使用表达芯片共检测到差异基因34个,其中下调表达的基因27个,上调表达的基因7个。在下调表达的基因中有7个(OAS1、IRG6、ISG20、DDX58、IFITl、ISG15、CXCL9)与干扰素诱导相关,1个(FST)与生长性状相关, 2个(MX1、PG-2)与抗病毒有关,2个(S100A9、Numb)与细胞生长和凋亡有关;上调表达的基因功能还不清楚,推测它们与提高机体的抗寒性有关。下一步拟计划采用高通量测序方法对冷应激相关基因进行筛选。

英文摘要: In order to analyze the cold resistance of Min pig, 12 piglet were divided into two groups randomly. One was feed outside (-12℃, the other was feed in house (15℃, after 24h, the serum of 12 piglet were collected. INS, EPI,T3 and T4 concentration were detected by ELISA. Some muscle tissues were collected, different expression genes were detected by Affymetrix expression chip. The results showed that, after 24h cold stress, the concentration of INS increased comparing with control group, EPI,T3 and T4 were all increased, but not significantly. Thirty-four differentially expressed genes were detected, among them, twenty-seven genes were down-regulated and seven were up-regulated. Among down-regulated genes, seven (OAS1, IRG6, ISG20, DDX58, IFITl, ISG15, CXCL9) were associated with interferon induction, one (FST) was associated with growth, two (MX1, PG-2) were associated with anti-virus, two (S100A9、Numb) were associated with cell growth and apoptosis. But the specific functions of the up-regulated genes were unknown, speculated they may have relationship to animal cold resistant.The next step is planned to use high-throughput sequencing method to find cold stress related genes.
