中文摘要: 鸭瘟病毒(DPV)可致水禽(鸭、鹅等)发生急性、败血性传染病,发病率和死亡率高,危害严重。本课题组获得了DPV中国强毒株的全基因序列(GenBank登录号JQ647509),然而目前对DPV基因功能知之甚少,特别缺乏关于DPV皮层蛋白与病毒成熟和免疫逃避关系研究。本课题以编码DPV皮层蛋白UL14为靶基因,通过构建基因缺失病毒和表达细胞系、免疫共沉淀、瞬时共表达、酵母双杂交、免疫电镜及病毒形态学观察等技术手段,探讨UL14蛋白与DPV其他相关蛋白(衣壳蛋白UL35、DNA包装蛋白UL33和调节蛋白UL48)之间的相互作用、UL14基因缺失病毒的形态学和毒力变化、UL14表达细胞系对细胞凋亡的抵抗等,从而解析UL14在病毒成熟和免疫逃避中的作用。研究结果将加深人类对DPV复制与繁殖过程的认识,为阐明DPV的致病机制提供科学数据,为防治DPV新药研究提供理论指导,具有重要科学价值和理论意义。

英文摘要: Duck Plague Virus(DPV) is a causative agent with an acute, septicaemia infection to waterfowl(duck, geese,et al). It has severely affected the waterfowl industry because of its high morbidity and mortality. Complete genomic sequence of Chinese virulent DPV was sequenced by our research group(GenBank accession number JQ647509). Little information about genes function from DPV was knows till now, especially about the relationship between tegument protein with virus mature and immune escape. Our application program is focus on encoded tegument protein UL14 gene, with the works of construction of UL14 gene deletion virus mutants and expression cell-line, immune co-precipitation, transient co-expression, yeast duplex hybridization, immune electron microscope and DPV morphology study et al. To approach the interactions between UL14 protein with other DPV-associated proteins(capsid protein UL35, DNA package protein UL33 and regulatory protein UL48), morphology and virulence of DPV-ULl4 gene deletion strains, anti-apoptotic of DPV-ULl4 gene expression cell-line , et al. Based above research works, we hope to explain how and why the DPV-ULl4 gene functional in virus mature and immune escape.
