中文摘要: 猪肌内脂肪含量是反映猪肉品质的主要指标,但对人类而言,脂肪在肌组织的过度沉积则可导致一系列疾病。因此,解析脂肪异位沉积的分子机制是改善肉用动物肉质品质及预防治疗人类肥胖相关疾病研究的焦点问题。项目组最近的研究表明,miR-34a参与了抵抗素介导的脂肪在肝脏及肌肉组织的积累,但其具体机制尚不清楚。本研究拟在前期研究工作的基础上,通过生物信息学分析miR-34a靶基因及调控信号通路,构建猪肝源和肌源MSCs成脂分化细胞模型,并通过细胞转染及活体水平研究miR-34a对动物脂肪异位沉积的影响,通过启动子双荧光素酶报告系统及ChIP、EMSA实验分析抵抗素对miR-34a调控的可能性机制,通过对miR-34a靶基因干扰及超表达分析其在动物组织脂肪形成中的作用,从而系统解析miR-34a参与抵抗素诱导的猪脂肪异位沉积分子机制。

英文摘要: Intramuscular fat (IMF) content of pocine is very important to the pork quality, which related with the visual appearance, smell, firmness, juiciness, tenderness and flavor. For human beings, however, the ectopic deposition of lipids in liver, muscle etc. may induce many diseases such as non-alcoholic fatty liver disease(NAFLD) and cardiovascular disease(CVD). So, it is of value to clarify the molecular mechanism of ectopic deposition of lipids to genetic improvement and strain cultivation of carcass and meat quality performance of animals, and also to prevention and treatment of diseases associated with obesity. Our recent experimental investigation shows that miR-34a involved in the development of ectopic deposition of lipids, however, the exact mechanisms underlying these processes remain unclear. In this study, we will try to analyse the target genes and pathways using bioinformatics technology, construct the inducible adipogenic differentiation cell model of pocine liver- and muscle-derived mesenchymal stem cesss,illustrate the influences of miR-34a on the formation of fat in liver and muscle tissues both in vivo and in vitro,find the regulatory pattern of resistin to the cis-acting elements and trans-acting factors on the miR-34a promoter through dual luciferase reporter system,chromatin immunopricipitati
