中文摘要: 博来霉素水解酶(BLH)因能水解博来霉素使其失效而得名,具有解毒、神经保护、免疫调节等多种功能。我们先期的研究发现:抑癌基因p53通过转录因子AP2能调节BLH的表达。但BLH如何与类泛素化结合酶UBC9相互作用,如何影响细胞应激反应,未进行深入研究。本项目使用多种细胞和分子生物学技术,通过对BLH缺失的细胞、神经细胞的研究,阐明BLH与UBC9相互作用的功能和对类泛素化的影响,反向调节p53稳定性的分子机制;确定BLH在DNA损伤反应、毒物兴奋效应以及细胞衰老中的作用机制及其信号通路。本项目的研究结果将进一步阐明BLH的生理与病理作用机制,为防治神经退行性疾病、心脑血管疾病提供理论依据和药物靶点。

英文摘要: Bleomycin hydrolase(BLH) is well known as it hydrolated bleomycin into inactive state. It plays several physiological roles, such as detoxinfication, neuron protection and immune-regulation. In our previous study, we have demonstrated that the BLH expression is regulated by p53 through transcription factor AP2. However, it remained unclear what function BLH binds sumorylation-conjugated enzyme UBC9 and how it effects stress responses. Using a lot of cell and molecular biological techniques, the present application will clarify the function of BLH and UBC9 interaction, effects on sumorylation and molecular mechanism of negative modulation of p53 stability in the BLH knock-out cells and neuron cells. It will also demonstrate the functional mechanism and signaling pathways of BLH in DNA damage response, hormesis and cellular senescence. The results obtained by this project will further elucidated the physiological and pathological roles of BLH and provide the evidence for prevention and treatment of degenerative neuron disease, cardiovascular and cerebrovascular diseases and drug target.
