中文摘要: 本项目基于适配子生物探针独特功能和上转换发光纳米晶(UCNPs)独特发光性质,构建基于UCNPs与TAMRA荧光猝灭分子(受体)间能量共振传递原理的均相免疫检测分析用新型生物光开关,探索解决当前荧光供体制约荧光标记适配子生物探针应用发展的关键科学问题。研究提高同质核/壳结构UCNPs发光上转换效率的有效新途径,获得性能优异的可用于标记适配子的供体。研究UCNPs表面生物偶联适配子的新方法和最佳物理参数,以保持适配子生物的光开关特异性和灵敏度。研究标记适配子的UCNPs、受体链和目标检测分子间相互作用动力学过程以及在近980nm红外光激发下,UCNPs供体与相应荧光猝灭基团 (如TAMRA)间LRET效率的影响。研究构建的UCNPs标记适配子生物探针性能在生物检测中的稳定性及其对靶分子竞争结合动力学过程,评价其生物检测特异性和灵敏度,为适配子在均相免疫检测分析中的应用提供新型供体和物理基础。

英文摘要: To solve the problem of the donors on the breakage and damage of aptamers LRET-based optical biosensor of an aptamer-upconversion conjugate was constructed base on the advantages of aptamer used for biodetection and the upconversion nanoparticles, which can perform infrared-to-visible photon conversion in the doped rare-earth ions. The research included: finding an effective way to improving the efficiency of upconverison luminescence; studing on the method, by which the nanoparticles could be bonded with avidin-biotin at 5'end of aptamer in order to keep their the high sensitivity and specificity; invesgating the energy transfer and dynamic process between UCNPs labelled aptamer and the quenching fluorescence TAMRA; finally evaluating the stability, sensitivity and specificity of the as-designed biosensor. This biophotonswitch has promising potential for applications in broad areas, such as homogeneous bioassay, liquid biochip or liquid array for bioassay and in situ imaging, where reusability and excellent photostability as provided by this biosensor, are greatly requested.
