中文摘要: 在时间和空间上调控特定基因的表达对基因功能的研究和新疗法的探索有着重要的意义。本项目拟合成四种光敏感的核苷亚磷酰胺单体和1-(2-硝基苯)乙醇的亚磷酰胺单体,通过DNA固相合成仪将其插入到siRNA单链序列的特定位置,分别合成在不同位点的磷酸二酯骨架单光敏基团修饰的siRNA的正义和反义链RNA,研究光敏基团修饰后的siRNA的稳定性和构象,以及在细胞水平上对外源和内源基因表达沉默活性的抑制以及光激活后siRNA基因沉默活性的恢复。找出影响光敏感siRNA沉默活性的若干关键修饰位点,使其既能够完全阻碍基因表达而光照释放光敏感基团后又能够使得siRNA的活性充分恢复,为研究特定基因的功能和致病基因在时空上选择性调控奠定基础。

英文摘要: Spatiotemporal regulation of gene expression is very important to gene fuction research and new gene therapy. We will synthesize four corresponding photoresponsive phosphoramidites from appropriate protected nucleosides and 1- (2- nitrobenzyl) ethanol phosphoramidite for RNA solid phase synthesis. The phosphoramidites are inserted to a particular location of sense strands and antisense strands of siRNA sequences by DNA solid-phase synthesizer. The photoresponsive modification at various positions on the phosphate backbones of siRNAs is done to investigate their stability, conformation and the ability to both knockdown endogenous and exogenous gene expression in cells and restore the normal level of activity upon irradiation. We hope to find key residues on the siRNAs with photolabile modification which maximizes the photomodulation of target gene expression. The ability will show promising application in spatiotemporal regualtion of specific gene function and disease genes.
