中文摘要: 本项目是国基金已结题项目的深度延伸。前期不仅发现产自贵州的土茯苓(Smilax glabra Roxb.)切面为红色,而且还发现切面红色与切面白色土茯苓的主要化学成分和药效有明显差异。为此,申请人拟以生物活性为导向对具有地域特点的切面红色土茯苓进行有效组分筛选,运用UV、IR、MS和NMR等波谱技术对其化合物进行结构鉴定,阐明效应物质,建立UPLC-PDA-MS标准指纹图谱,以期实现用效应物质来控制药材质量;继而建立大鼠佐剂关节炎模型(AA),从表征观察、细胞因子的表达与mRNA的转录水平、关节病理等宏观与微观两方面,寻找切面红色土茯苓对类风湿性关节炎(RA)干预作用的重要依据。研究结果不仅可阐明切面红色土茯苓对RA干预作用的效应物质及作用机制,还可为下一步开发贵州药用资源、新产品及获取地理标志的保护奠定基础,也可为法定标准的补充提供可能。

英文摘要: This is an extension project of previous fund Natural Science Foundation of China (NSFC) program, in which, Smilax glabra is classified as white or red species based on the color of cross section. The red one is mainly produced in Guizhou and we found that both main chemical compositions and pharmacological effects between these two species are significant different. In this project, the effective substance of red-Smilax glabra(red-SG) will be screened based on its biological activity, the structural of its chemical compounds will be identified by UV, IR, MS, NMR etc, and its standard fingerprint will be established by means of UPLC-PDA-MS for the quality control of medical material. In addition, the intervention effect of red-SG on rheumatoid arthritis (RA) will be verified by characterization observation and joint pathology as well as evaluation of cytokines expression and their mRNA transcriptional level from macroscopic and microscopic aspects. The outcomes of this study are beneficial to further identify the effective substance of red-SG, to reveal its mechanism of the intervention effect on RA, and therefore contribute to develop new resource and product as well as provide new evidences for complement of legal standards and protection of geographical indication.
