中文摘要: 炎症对机体有损伤作用,甚至导致癌症、糖尿病和心血管等重大疑难疾病。Beta-葡聚糖显示抗炎症活性,但由于它的链结构复杂、形态多样,导致其链结构-抗炎症活性构效关系及其与靶标分子间的相互作用研究报道很少。本项目拟采用高分子物理理论和方法系统研究不同来源的药用植物或菌类细胞壁beta-葡聚糖及其衍生物的链结构(键接方式、支化、梳型、分子量、链尺寸、螺旋链、无规线团等),解析导致结构相似或相同而链形态各异的分子内、分子间的相互作用;同时,结合细胞和分子生物学方法研究它们的抗炎症活性,揭示其链结构-抗炎症构效关系;用SPR、CD、QCM、荧光等研究beta-葡聚糖与各种潜在靶标受体分子间的相互作用和分子识别机制,并结合基因敲除技术进一步确定其靶标受体分子,揭示链结构影响受体分子识别的科学规律。由此,本项目将为研究和开发多糖类抗炎症新药提供科学基础,具有重要学术价值和应用前景。

英文摘要: Inflammation is harmful to the body, even leading to cancer, diabetes, cardiovascular disease, and so forth. Beta-glucan shows anti-inflammatory activity, but few reports are visible on its chain structure-antiinflammation correlation and the interactions with its target receptors due to the complexity of structures and chain shapes of bata-glucans. This project will investigate the chain structures including linkage,branche, combtype, molecular weight, chain size, helical chain, and random coils etc. of beta-glucans from cell walls of medicinal palnts or fungi and its derivatives by using polymer physics theories and methods, and analyze the intra- and inter-molecular interactions between the macromolecues of beta-glucan which result in the various chain shapes; simultaneously, the anti-inflammatory activities will be determined by using celluar and molecular biology technics, revealing the relationship between the chain structure and anti-inflammation of beta-glucans; the binding ability of beta-glucan to its possible receptors and its molecular mechanism will be evaluated by SPR, CD, and QCM etc.; the final target molecule will be further confirmed by gene knockout technology, and the scientifical law by which the chain structures affect the recognition of beta-glucans by the target receptors will be propose
