中文摘要: 课题以导致果汁混浊的主要成分槲皮素-3-鼠李糖苷(H3S)、山奈素-3-鼠李糖苷(S3S)、山奈素-3-芸香糖苷(S3Y) 和糖蛋白Fr1为研究对象。NMR获得Fr1空间结构基础上进行计算机模拟,阐明H3S、S3S、S3Y与Fr1相互作用部位氨基酸残基的组成以及相互作用部位空间结构;对溶液体系中H3S、S3S、S3Y与Fr1相互作用的结合常数,结合位点数及相互作用力荧光光谱研究基础上,相互作用对溶液中Fr1构象影响的紫外可见光谱、拉曼光谱和圆二色谱研究的基础上,Biacore 3000实时检测分子间的相互作用;多功能扫描探针显微镜、激光共聚焦扫描显微镜和激光粒度分析仪测定从纳米到微米再到毫米尺度复合物的动态变化,揭示相互作用处结构域与果汁形成混浊之间的关系,指导筛选全天然食品添加剂定点封闭糖蛋白特定结构域,阻止天然果汁中糖蛋白与多酚的相互作用来防止混浊的形成,使果汁中保留更多多酚和糖蛋白。

英文摘要: Quercetin-3-rhamnoside(H3S),Kaempferide-3-rhamnoside(S3S),Kaempferide-3-rutinoside(S3Y) and glycoprotein (Fr1) which were the main compositions for the haze formation in juice is the object of this study. On the basis of glycoprotein spatial structure obtained by NMR , computer simulation was applied for revealing the three-dimensional structure and amino acid residues sequence of the interaction domain between H3S, S3S , S3Y and Fr1. The binding constant, number of binding sites and the interaction force between H3S,S3S,S3Y and Fr1 in solution were investigated by fluorescence spectra, the effects of interaction on Fr1 conformation were also detected by UV-visible spectroscopy, raman spectroscopy and circular dichroism. Based on this results, the interaction between moleculars were studied real -time by Biacore 3000. The dynamic changes of complexes obtained by the interaction between H3S,S3S,S3Y and Fr1 from nanometer to micron and then to millimeter were detected by multifunctional scanning probe microscopy, confocal laser scanning microscopy and laser particle size analyzer.
