中文摘要: 除草剂对于保证农业生产具有重要的作用,随着杂草抗性的增加和环保要求,迫切需要开发高效、低毒和环境友好的新型除草剂。由于目前已知的除草剂作用机制和靶标数量有限,已成为除草剂开发的瓶颈,因此寻找新型的作用靶标以及解析靶标蛋白的结构则成为新农药开发的首要条件。本项目根据新型除草剂开发的实际需求,借鉴医药开发的成功经验,利用已获得的模式生物拟南芥和水稻的基因组序列,采用序列比对等生物信息学方法筛选出具有潜在除草剂作用靶标价值的蛋白激酶基因。结合结构基因组学的研究方法和技术对确定的靶标基因进行大规模的克隆、表达和纯化,并采用多维核磁共振波谱技术和X-射线晶体学方法解析相关蛋白质的三维空间结构,为基于靶标结构的除草剂分子合理设计提供理论和结构基础,从而为新型靶标的选择和研究提供新的方法,促进我国新型除草剂创制的源头创新。

英文摘要: Herbicide plays an important role in agricultural production. With the increase of weeds resistance and the environmental protection request, it has an urgent need to develop more effective, low toxicity and environmental friendly new herbicides. Because the present known herbicides mechanism and targets are limited, it has become the bottleneck of herbicide development, so searching for new targets and determination of the target protein structures are the primary stage of target structure-base new herbicide design and development. According to the actual demand of new herbicide, we propose a structural genomic study on novel potential protein kinase-based herbicide targets. Refer to the successful experience of medicine development, new kinase genes from Arabidopsis thaliana and Oryza sativa genome will be functional analyzed by comparative genomics method, potential herbicide target genes will be cloned, expressed with structural genomics techniques. The three-dimensional structures of soluble proteins will be determined using multidimensional magnetic resonance spectroscopy and X-ray crystallographic technology, respectively. The present study would provide reasonable herbicide mechanism and basic structures of new targets, and promote the novel herbicides development innovation.
