中文摘要: 细菌中氨基酸分泌蛋白的适度表达,能够显著提高氨基酸的细胞外积累量。谷氨酸棒杆菌是工业发酵生产氨基酸的重要菌株,但是目前在该菌中仅识别出四个氨基酸分泌蛋白,这与分泌蛋白识别方法单一,依赖于特异性分泌蛋白基因缺失突变株有关。本项目拟结合现代生物信息学分析/比对技术、氨基酸分泌蛋白的快速筛选方法和基因敲除/互补方法直接进行谷氨酸棒杆菌中氨基酸分泌蛋白的预测和鉴定,并从分子水平上揭示谷氨酸棒杆菌中新氨基酸分泌蛋白的生物学特征和调控机理。研究组前期通过先预测再鉴定的方法,从谷氨酸棒杆菌中识别出两个氨基酸内运蛋白,为本项目的顺利进行提供了一定的技术支持。本项目的研究对于建立新的氨基酸分泌蛋白识别方法,认知谷氨酸棒杆菌氨基酸分泌蛋白及其调控机理,以期应用于生产菌株改造具有重要意义。

英文摘要: The moderate expression of amino acid exporter in bacteria can significantly raise the amino acid extracellular accumulation level. Corynebacterium glutamicum is widely used for the biotechnological production of amino acids, but at present only four amino acids exporters have been identified in Corynebacterium glutamicum. It is associated with the single recognition method and the dependence on specific amino acid exporter auxotroph. The project will establish a rapid screening method of amino acid exporter, combining with modern bioinformatics analysis/alignment and gene knockout/complementary method to predict and identify new amino acid exporters, and reveal them molecular characteristics and regulation mechanism in Corynebacterium glutamicum. The group has identified two amino acid importers by predict-screening method, provided techincal support for this project. The project is helpful to establish a new identification method of amino acid exporter, to cognize amino acid exporters and their regulations and to improve production strains.
