中文摘要: 癌症,被称为人类健康"第一杀手",严重威胁着人们的生命安全。癌症的早期发现、早期诊断及早期治疗是降低死亡率及提高生存率最有效的手段。对肿瘤标志物进行检测,可以为肿瘤早期诊断,检测肿瘤复发与转移,判断疗效及预后等提供重要依据。然而现有肿瘤标志物检测方法多有灵敏度不足、易出现假阳性等局限。本项目利用核酸探针的优良特性,提出将自组装DNA串联体作为信号放大载体的新方法。前期研究表明,在优化条件下,自组装DNA串联体的长度超过20微米,可负载大量的电化学标记物,从而产生明显放大的电化学信号。基于此,本项目拟以临床常用的若干特异性肿瘤标志物为研究对象,对该方法进行更系统、更深入的研究,期望建立一种超灵敏、高选择性、更简便、更快速的信号放大的共性方法,并将此方法应用于生物样品中多种超低含量肿瘤标志物的同时检测。此方法的建立及其在生物医学领域的应用,具有重要的研究价值和实际意义。

英文摘要: Cancer, a serious threat to the safety of people's life, is named as the first killer of human health. The most effective means to reduce mortality and improve survival is try to achieve early detection, early diagnosis and early treatment of cancer. Tumor marker detection can provide an important basis for early diagnosis of tumor, early detection of tumor recurrence and metastasis, and evaluation of treatment efficacy and prognosis. However, existing methods for the detection of tumor markers have many limitations, such as low sensitivity and false positive. Based on the properties of the nucleic acids, we propose a new method using self-assembled DNA concatamers as carriers for signal amplification. Preliminary studies show that, under optimized conditions, the self-assembled DNA concatamers longer than 20 μm can accumulate many electrochemical indicators and produce a much larger signal. In order to study the method more systematically and thoroughly, some specific tumor markers commonly used in clinical trials are chosen as the subjects investigated. We try to establish a common method for signal amplification.
