中文摘要: 膜联蛋白(Annexin)家族是一种钙依赖性的结合磷脂和微丝的蛋白,参与许多植物重要的生理活动。已有的报道表明,膜联蛋白定位于花粉管中,我们推测该家族成员可能是一种潜在的联接膜、微丝骨架和钙之间的 "枢纽"蛋白,在花粉管极性生长过程中起着极其重要的作用。然而,目前尚未见该家族成员参与调控花粉管生长的报道。因此,本项目拟采用分子生物学、遗传学和细胞生物学等方法,首次尝试研究拟南芥花粉特异表达的该家族成员-annexin5的器官组织和亚细胞定位;annexin5在体外对微丝聚合和解聚的调控作用,以及与其它微丝结合蛋白和囊泡的相互作用关系;annexin5对烟草或者拟南芥花粉管和拟南芥植株生长的影响作用,旨在揭示植物膜联蛋白参与调控花粉管极性生长的分子机制。本项目研究成果有可能为植物细胞极性生长机制、植物信号传导、植物抗逆以及作物杂交育种等相关研究提供新知识、新材料和新方法,具有非常重要的理论研究意义和实际应用价值。

英文摘要: Annexins belong to a multifunctional family of calcium-dependent, membrane and F-actin binding proteins, which are localized in the tip of the pollen tube. We deduced that annexin family proteins might be the ideal mediators in cell membrane, calcium and actin cytoskeleton interactions, and play a crucial role in regulating the pollen tube growth. However, the function of annexin in regulating the pollen tube growth has not been reported yet. Here we aim to study the organic and cellular localization of annexin 5 which is expressed specifically in arabidopsis pollen, as well as the function of annexin 5 in regulating the actin polymerization and depolymerization in vitro, the reaction of annexin 5 with other actin binding proteins and vesicles, and the effect of annexin 5 in regulating the growth of tobacco or arabidopsis pollen by using the method of molecular biology and cell biology. Our research may illuminate the molecular mechanism of the annexin regulating the pollen tube growth. This project may provide the new infomations, materials and methods in the field of plant cell polar growth, signal transduction, plant breeding and so on. The results are very important for the oretical research and worht for practical application.
