中文摘要: 抑郁症是严重危害人类健康的重大精神疾病,神经网络的异常改变是其物质基础,采用结构及功能影像学研究抑郁症相关脑网络异常已成为新的热点。尽管抑郁症患者影像学研究已初步构建了情感障碍的众多病理环路假说,但因人体试验的伦理学限制,难以采用其他方法进一步验证。因此,采用合适的动物模型研究抑郁症情感环路关键节点的功能及其网络关系意义重大。 本项目拟在前期构建食蟹猴抑郁模型的基础上,采用7T超高场强磁共振技术观察正常-抑郁-恢复过程中神经网络的动态改变,结合三维偏振光纤维成像技术,构建猴抑郁行为相关网络模型;在此基础上,采用电刺激或损毁实验,结合行为学反应,深入验证该模型关键节点的功能及各节点间交互影响和调控的内在机制,为抑郁症这一重大疾病发病机制的深入探讨提供依据,也为后续情感环路的进一步生物学研究奠定基础。

英文摘要: Major Depression (MD) is a serious hazard to human health. Abnormal changes in the neural network is the key basis of MD. Structural and functional neuroimaging studies of depression-related brain networks has become a new hot spot. Previous neuroimaging studies have offered various hypothesis of pathological loops of affective disorders, but it is difficult to use other methods to further verify the contradictory results. Therefore, the use of a suitable animal model of depression is the key to study interactions of different nodes of the related network. This project intends to build a monkey model of depression, and to use 7T ultra-high field MRI to dynamically observe the neural network changes during the healthy-depressed-rehabilitation process, combined with three-dimensional polarized light fiber imaging technology, to build the core network related to monkey depressive behavior. On this basis, functions of key nodes in the network such as the amygdala, medial prefrontal cortex will be explored using electrical stimulation (or damage) methods. This project will not only provide new insight for understanding the mechanism of major depression, but also lay the foundation for further biological studies of the related emotional network.
