中文摘要: 颅内海绵状血管瘤(CCM)是常见的颅内血管畸形,由于其出血率高、预后差,对患者生活及经济产生极大的负担。阐明CCM的血管形成机制将有助于减低疾病的风险,谋求除手术及放疗之外新的干预措施即血管抑制治疗。我们在前期研究中发现CCM患者的血管内皮细胞中存在CHI3L1基因的过度表达现象,而最新研究提示CHI3L1的转录蛋白YKL-40具有促进血管生成的作用。因此,本课题拟通过多种手段检测CCM组织中YKL-40的表达水平及差异;构建CHI3L1差异性表达的慢病毒载体及外源性YKL-40添加,在细胞水平观察血管内皮细胞功能的改变;进一步通过体内实验观察及验证局部CHI3L1不同表达环境对血管形成的影响。通过深入探讨CHI3L1基因和YKL-40在CCM组织中的表达及其对CCM血管形成的调控作用,阐明CCM血管发生发展的细胞与分子生物学机制,为寻求诊断及治疗CCM的新方法提供可靠的理论依据。

英文摘要: Intracranial cavernous malformation(CCM) is commmon in intracranial vascular malformation. Patients with CCM was threatened by its high haemorrhage risk and poor prognosis. It will help find a new theraputic method excepted from neurosurgery and radiosurgery to improve the prognosis by identify the pathologic angiogenesis mechanism. Our primary study showed CHI3L1 gene was overexpressed in CCM vascular endothelial cells. Recent studies also indicated that YKL-40 protein (transcripted by CHI3L1) participated in the process of angiogenesis. Thus, based on the examination of YKL-40 expression in CCM tissue through various methods, our group plan to further investigate and illustrate the regulation pattern of YKL-40 in the angiogenesis process of CCM though the in vitro cell migration study, 3D matrigel tube formation assay and in vivo mice CCM model establishment. The result of our study may theoretically support to explore a new diagnositic and htheraputic way in CCM.
