中文摘要: 脱氧雪腐镰刀菌烯(DON)是目前粮食与饲料中污染较严重的真菌毒素之一,易造成人和动物的急慢性中毒。如何预防或降低摄入DON后造成的急慢性中毒的风险,是一个严峻而又亟待解决的科学问题。从分子作用机制来看,p38 MAPK是一条应激信号通路,也是唯一的DON与S.boulardii作用细胞的相同信号通路。S. Boulardii能抑制外源因子对此信号通道的激活,抑制相关炎性细胞因子表达,减轻炎症反应;DON导致细胞毒性则是通过激活p38 MAPK细胞信号通道,促使炎性细胞因子的表达,但两者同时作用细胞时,如何通过该通路调控细胞凋亡的作用机制尚未明了。本项目利用猪单核巨噬细胞为研究对象,通过信号通路抑制剂、免疫印迹、流式细胞术、RT-PCR等技术手段,研究p38MAPK在S.boulardii对DON所致猪单核巨噬细胞凋亡的干预作用机制,为该菌作为防治DON导致的急慢中毒的药物奠定理论基础。

英文摘要: Deoxynivalenol(DON)naturally contaminated foods and feeds animal feeds, is toxic and carcinogenic to humans and animals. How to prevent or reduce the risk of DON has been emerged as a big scientific problem. Viewing from molecular mechanism, protein kinases (MAP kinases) p38 signaling pathways is not only an important stress pathway but also the same target for DON and S.boulardii. The main mechanisms of action of S. boulardii inhibits production of pro-inflammatory cytokines MAP kinases, p38 by inhibiting main regulators of inflammation; in contrast, DON evokes production of MAPK p38 to induce inflammation. Whereas it remains unknown how they simultaneously regulate cell apoptosis through that same pathway. For the purpose of better understanding the role of MAPK p38 on S.boulardii reducing the acute or chronic poisoning caused by DON, this research is developed in pig's mononuclear macrophages apoptosis caused by DON ,and used a series of technologies such as the signal pathway inhibitor, immune imprinting, flow cytometry, RT-PCR to explore the intervention mechanism of S.boulardii to pig's mononuclear macrophages, which would be helpful for lay a theoretical foundation to explore the drug preventing DON.
