中文摘要: 证候生物学基础是中医学有待突破的关键科学问题之一。本课题基于既往研究基础和探索,提出证(四诊信息群)存在相对应的'代谢物质组合群(谱)'的假说。 拟以非酒精性脂肪肝(NAFLD)典型的肝胆湿热证和肝郁脾虚证患者为研究对象,以NAFLD隐证型(无证可辨)患者及健康志愿者为对照,在四诊信息收集基础上,运用 GC/MS和LC/MS技术进行尿代谢组学检测分析,获得不同典型证候的尿特征性代谢物。运用证候数理模型分析技术分别建立中医证候信息的表达模型及与之相应的代谢物质组合群(谱)。在此基础上,选择典型的肝胆湿热证患者,以临床用于NAFLD肝胆湿热证的胆宁片作为干预的载体,进行干预前后代谢物质组合群(谱)变化与证候变化的关联性分析,验证代谢物质组合群(谱)的有效性,以期论证申请者提出的假说,从而揭示中医证的生物学基础,并为证的客观评价奠定方法学基础。

英文摘要: Syndrome biological basis is one of the key scientific problems needed to be broken in traditional Chinese medicine. Based on previous study foundation and exploration, this study put forward hypothesis that syndromes (four diagnostic Information groups) exist corresponding to metabolites combination groups (spectrum). Typical gallbladder damp-heat syndrome and liver depression and spleen deficiency syndrome in patients with non-alcoholic fatty live disease (NAFLD) are treated as study subjects, occult syndromes(no symptoms for analysis) in patients with NAFLD and healthy volunteers are treated as control group, on the basis of four diagnostic information collection, urine metabonomic studies will be carried out with mass spectrometry (MS) combined with gas chromatography(GC/MS),liquid chromatography (LC/MS), urinary characteristic metabolic substances of different typical syndromes will be obtained. Syndromes information expression model and its corresponding to metabolic substance combination groups (spectrum) will be established by using syndrome mathematical models analysis techniques. On this basis, typical gallbladder damp-heat syndrome patients will be selected, danning tablet for the treatment of liver and gallbladder damp-heat syndrome of NAFLD will be treated as a carrier intervention, correlation analysis will be carried out betweent alteration of metabolic substance combination groups (spectrum) and syndromes changes before and after the intervention,and verify the validity of metabolic substance combination groups (spectrum). Hypothesis put forward by the applicant will be demonstrated ,and thus reveal the biological basis of TCM, and lay methodology foundation for syndrome objective evaluation.
