中文摘要: 多囊卵巢综合征(PCOS)中胰岛素抵抗是主要的发病机制之一,我们的前期工作中证实PCOS患者的核心家系中也存在胰岛素抵抗。但目前对PCOS胰岛素抵抗发病机制的研究多集中于特定的基因突变及转录异常,缺乏对相关信号转导途径的基因调控机制的深入研究。本项目拟采用miRNA芯片技术构建PCOS抵抗的miRNA差异表达谱,筛选PCOS胰岛素抵抗相关miRNA;应用生物信息学技术预测PI3K/Akt及其上、下游基因相关miRNA;应用双荧光素酶报告基因检测系统进行实验验证,证实其调控作用。

英文摘要: Insulin resistance plays an important role in polytcystic ovary syndrome(PCOS) pathogenesis. Our former research also confirmed there was insulin resistance in the PCOS trios. It is unclear about the regulation mechaninsm of insulin signaling pathway since the most researches concentrated on gene mutation and abnormal transcription. Our aims in this study are to use miRNAs chip to evaluate if insulin resistance in PCOS. And miRNA related to insulin resistance will be selected. Bioinformations will be used to find the possible miRNA targeting sinaling pathway PI3K/Akt and IRS/Glut. The probable miRNA targeting PI3K/Akt will be verified by dual luciferase reporter assay.
