中文摘要: miRNA是一类真核生物内源性小分子单链RNA,通过引发靶mRNA的降解或翻译抑制来介导转录后基因沉默。除多细胞生物外,研究发现病毒也能够编码miRNAs,并可通过作用于病毒或宿主基因,在病毒感染复制及阻断宿主免疫监控中起重要作用。我们的前期工作证实,海洋鱼类病毒也可编码miRNAs,通过Solexa深度测序共鉴定得到石斑鱼虹彩病毒SGIV编码16个miRNAs,其中11个具有生物学活性,但其在病毒感染过程中的作用尚不清楚。基因组定位提示,具生物学活性的miRNAs中,miR-homoHSV的潜在靶基因是SGIV编码的凋亡相关蛋白-脂多糖诱导的肿瘤坏死因子类似物LITAF。本课题拟在前期工作基础上,深入研究miR-homoHSV在病毒感染复制以及病毒诱导程序性细胞死亡过程中的调节作用。该项目将为阐明鱼类病毒感染及致病的分子机理提供重要信息,并为研究海洋鱼类病毒防治手段提供理论依据。

英文摘要: miRNAs are a class of small noncoding RNA in eukaryote that post-transcriptionally regulate gene expression by cleavage of the target mRNA or translational repression of the mRNA transcripts. Besides metazoan eukaryote, viruses were found to be able to encode miRNAs. Virus encoded miRNAs can target special genes of virus or/and host in order to facilitate viral replication and also evade host immune responses. Our previous study demonstrated that marine fish virus can encode miRNAs, and 16 novel miRNAs encoded by Singapore grouper iridovirus were identified by Illumina/Solexa deep-sequencing technology, while 11 of these viral miRNAs were present and functional in SGIV-infected host cells. To date, the roles of SGIV miRNAs in regulating virus replication and virus-host interaction remain largely unknown. miRNA localization indicates that, the potential target of miR-homoHSV among the 11 functional SGIV miRNAs, is SGIV encoded lipopolysaccharide-induced TNF-a factor which is related to apoptosis. For this project, we will investigate the roles that miR-homoHSV plays in SGIV infection, especially its modulation on SGIV induced programmed cell death (PCD).
