中文摘要: 骨质疏松性骨折愈合过程缓慢,恢复时间长,易发生骨折延迟愈合甚至不愈合,从而导致患者再次手术可能、康复时间延长、痛苦增加及经济负担增加。因此,寻找一种经济、安全、高效的促进骨质疏松性骨折愈合的治疗方法成为目前研究的热点,对提高人们健康水平具有重要的意义。肌肉微环境,是目前促进骨质疏松性骨折愈合的一条研究新途径。 本项目利用基因敲除、细胞转染和RANi等现代技术,从β-Catenin信号通路调节肌肉微环境中肌源性干细胞(MDSCs)成骨分化功能方面研究骨质疏松性骨折愈合的分子机理,并深入研究健脾补肾方动员MDSCs和促进其成骨定向分化的作用和分子机制,阐明健脾补肾中药介导β-Catenin信号通路调控MDSCs治疗骨质疏松性骨折的有效性和科学性,为中药联合肌肉干细胞治疗运用于临床奠定实验基础,丰富中医脾肾相关理论的科学内涵。

英文摘要: Osteoporotic fracture healing process is slow with long recovery time, and easy to happen delayed union,even nonunion of fractures, resulting in that the patient may take an operation again, with long recovery time, increasing the economic burden and pain. Therefore, looking for a kind of economic, safe, effective treatment for osteoporotic fracture healing has become the focus of current research, which has an important significance to improve people's health level. At recent, muscle microenvironment, has become a new way to promote the healing of osteoporotic fracture. In this project,using gene knockout, cell transfection and RANi and other modern technology, the molecular mechanism of osteoporotic fracture healing will be studied from the aspect ofβ-Catenin signaling in regulating the muscle-derived stem cells(MDSCs) to do osteoblastic differentiation. We will study that Jianpi-Bushen formulas mobilizes MDSCs and promotes its function to osteoblastic differentiation. We also will clarify that the regulation of MDSCs in treatment of osteoporotic fractures with Jianpi-Bushen formulas is effective and scientific, and provide experimental basis for clinical treatment of Traditional Chinese Medicine combined with muscle stem cell, and to enrich the scientific conntation of spleen and kidney-theory in Traditional Chinese Medicine.
