中文摘要: 骨髓间充质干细胞(MSCs)是修复损伤心肌的常用干细胞,但其在缺血缺氧微环境下的低存活率已成为MSCs临床应用的瓶颈。Netrin-1是一种神经介导蛋白,近年申请人研究其在心血管方面的重要作用, 发现Netrin-1可以促进心肌和内皮细胞NO的分泌,在体外心肌梗塞模型中保护心脏,而且还发现Netrin-1对MSCs的增殖和迁移有促进效果。本项目拟在前期研究的基础上,使MSCs过表达Netrin-1基因,检测Netrin-1的表达含量以及MSCs的增殖和迁移能力,评估Netrin-1对MSCs的影响。注射Netrin-1-MSCs到大鼠心肌梗塞部位,检测MSCs的定位分布及体内分化情况。对过表达的Netrin-1对改善MSCs的存活率,提高其增殖量、NO分泌量、改善心肌坏死面积及心脏功能等方面进行研究。可为临床应用MSCs治疗缺血性心脏病提供依据。

英文摘要: Mesenchymal stem cells (MSCs) transplatation therapy is used for decrease and repairment of heart injury. However it becomes a serious problem that low survival rate of MSCs in the hypoxia condition when the patient go through the ischemia reperfusion. Netrin-1 is a protein involved in axon guidance. Recently, applicant work on the netrin-1 function on heart protection, we discover that netrin-1 potently protects the heart from ischemia-reperfusion injury by stimulating NO production from cardiac ECs and myocytes. Moreover netrin-1 improve cell proliferation and stimulate angiogenesis. Therefore in the present study we aimed to investigate the therapy effect of netrin-1 overexpression MSCs on ischemia-reperfusion hearts. Netrin-1 expression, NO expression and cell proliferation will be examined. Netrin-1-MSCs will be injected into the infarted area of heart on rat model. Location, proliferation, differentiation and NO expression of MSCs and infart size and function of the heart will be examined. This study will provide a new potential targets and suggest novel therapeutic opportunities on managing ischemic heart disease may prove highly rewarding.
