中文摘要: Cpne5是个新的神经发育相关蛋白,主要表达于发育中的脑组织,其生物学功能尚不明确。我们近期研究表明:1)敲低皮层室区细胞Cpne5表达能显著促进新生神经元的放射状迁移,而过表达Cpne5能显著抑制神经元的放射状迁移;2)改变Cpne5表达能显著改变神经细胞形态;3)共定位结果显示Cpne5可能与细胞骨架调节分子MRCK相互作用。上述结果提示Cpne5可能通过与MRCK相互作用而影响细胞骨架的重排,从而在神经元放射状迁移过程中发挥负调控作用。因此,本研究旨在1:利用基因敲除小鼠验证Cpne5在神经元放射状迁移过程中的负调控作用;2:明确Cpne5负调控作用的发挥是否依赖MRCK介导的细胞骨架重排。本研究的结果有可能为神经元迁移异常导致的发育性脑病研究提供新的线索。

英文摘要: Cpne5 is a new neurodevelopment related protein that mainly expressed in the developing brain with no exact functions identified yet. Recently, our results demonstrated that inhibiting Cpne5 expression would significantly increase the number of neuron migrating to the cortex surface, and overexpressed Cpne5 protein could significantly decrease the number of neuron migrating to the cortex surface. Additionally, changing Cpne5 expression can change the neuronal morphology, and overexpressed Cpne5 can co-localize with MRCK in Hela cells. These results indicates that, Cpne5 may regulate the neuronal migration by affect cytoskeleton rearrangement. Thus, using Cpne5 knock out model, we plan to verify the role of Cpne5 in the radial neuronal migration and investigate the molecular mechanism of this process. The results of this project will help to study new mechanism of developmental brain disease caused by abnormal neuronal migration.
