中文摘要: 随着荧光识别及检测技术的不断发展,利用近红外荧光探针对于一些重要生物小分子的识别研究逐渐成为近期的研究重点。但是目前基于核苷酸分子识别的近红外荧光探针研究中,普遍存在信号响应时结构相似化合物间干扰严重,自主设计的新结构荧光团少及生物活体测试研究不够深入等问题。因此,我们计划设计合成4-6种新型基于花菁荧光团及苯并吡喃荧光团对核苷酸具有选择性识别功能的近红外荧光传感器,实现专一性强,灵敏度高,检测极限低的检测目标;并计划自主设计合成1-2种新型近红外荧光团,在此基础上制备3-5种对核苷酸具有选择性识别功能的近红外荧光探针,力争在分子结构设计及性能上有所突破创新;深入开展该系列近红外荧光传感器分子的生物活体成像应用研究,实现生物体活体荧光成像及全过程动态成像检测。

英文摘要: With the development of fluorescent detecting techniques, the research of near-infrared fluorescence sensing and fluorescence imaging for some important biology small moleculars has been an active research are in recent years. For the fluorescence nucleotide sensors, the problems, such as interferences from the moleculars with similar structures, difficulties of the design of new fluorophores and limits of the biological applications in vivo, can not be ignored. Therefore, we plane to develop 4-6 near-infrared fluorescent nucleotide sensors based on cyanine or dicyanomethylene-4H-chromene with high sensitivity and high selectivity. Meanwhile, we plan to design 1-2 new near-infrared fluorophores and 3-5 derivatives based on them. Based on the single-cell fluorescence recording, the exploration of the biology applications of these fluorescence sensors to realize the records in vivo and the dynamic imaging are our goals. Through all these efforts, we wish we could promote the developments of the research of near-infrared fluorescence sensing and near-infrared fluorescence imaging.
