中文摘要: 光老化机制的研究,细胞内酶及信号通路的相互作用(cross-talk)近年来已经成为最新的研究热点之一。我们的前期研究不仅与国外学者同期发现和证实了发现一种新型的组织蛋白酶CatG与皮肤光老化机制密切相关,近期的研究还提示其与MMPS蛋白酶、TGF-β/Smad信号通路有相互作用,通过共同影响I型胶原蛋白代谢参与皮肤光老化发生,但至今为止具体的机制尚不清楚。本研究拟用Anti-Cy3、(COL-1)ELISA、pSUPER、Anti-FITC等技术,研究光老化皮肤细胞内Cat G、MMPS与TGF-β/Smad信号通路相互作用的具体分子机制,及其在光老化真皮I型胶原蛋白合成/分解失衡中的作用。该研究不仅可进一步揭示光老化和日光相关性疾病的发生机理,使我们的研究紧跟国际前沿,而且可为光老化及相关皮肤病的防治提供新的靶点和思路,有十分重要的理论价值和实用意义。

英文摘要: MMPRIntracellular enzyme and signal pathway interaction (cross-talk) is a key point in skin photoaging mechanisms and it has become a forefront hot topic in this field. Our previous studies and recent literatures have indicated that CatG play a important role in skin photoaging process and may interact with MMPS and TGF-β/Smad pathway in UV-induced collagen-I synthesis and decomposition, however its underlying cytobiologic and molecular-biologic pathway remain unclear so far. This study mainly focus on investigating the interaction (cross-talk) of Cat G, MMPS and TGF-β/Smad pathway as well as collagen-I metabolism changes by utilizing Anti-Cy3、(COL-1)ELISA、pSUPER and Anti-FITC in photoaged cell and artificial skin model. The result of this study may help in expanding a knowledge of photoaging mechanisms and benefit the development of new anti-photoaging drugs and chemicals in further research.
