中文摘要: 近年来,C-H键活化直接形成C-C和C-杂键反应,由于其原子经济性、可持续性、环境友好等特征,越来越受到人们的关注。因为许多药物分子中涉及了唑杂环与碳、杂原子之间的连接,所以研究唑杂环的C-X(C、O、S、N等)键形成反应具有重要的意义。本项目利用唑杂环2-位碳易发生脱氢偶联反应的特点,研究了它和醛基、羟基发生C-H键活化/形成C-C键、C-O键的方法。进一步拓展了唑杂环的C-H键活化研究。课题一方面研究了醛基、羟基和唑杂环的分子内反应,构建了系列多样性稠杂环,为合成具有这些稠杂环的药物及其它生物活性分子提供了一条新的途径。另一方面还研究了醛、醇、酚、羧酸和唑杂环的分子间反应,为唑杂环,如苯并噁唑、苯并噻唑、N-取代苯并咪唑、噁唑、噻唑等的2-位碳引入酰基、烷(芳)氧基、酰氧基,建立了新的合成方法。

英文摘要: The C-H activation strategy for C-C and C-heteroatom bond formation has received substantial attention because of its economic, sustainable and environmentally benign features. Many pharmaceuticals contain heterocycle -carbon or heterocycle-heteroatom linkages, so the research of azole C-X (C, O, S, N, etc.) bond-forming reaction has important significance.In our project, using azole C-2 prone dehydrogenative coupling chatacteristics, we will study metal-catalyzed C-H activation/C-C(C-O)formation reaction between aldehyde,hydroxy and azole. First,we will develop intramolecule reaction between aldehyde, hydroxyl and azole, to build a series of diversity fused heterocyclic which provide a new approach for the synthesis of these fused heterocyclic drugs and other biologically active molecules. The other hand, we will also study the intermolecule reaction between the aldehydes, alcohols, phenols, carboxylic acids, and azole heterocycle, such as benzoxazole, benzothiazole, N-substituted benzimidazole, oxazole, thiazole and so on.It will establish new synthetic methods for the introduction of acyl, alkoxy (aryloxy) alkoxy, acyloxy into 2-position carbon of azole.
