中文摘要: 本实验室研究大鼠肝再生中肝细胞增殖调控发现,SPINK3与肝细胞增殖密切相关,然而其调控机制尚不清楚。为此,本项目拟进一步研究SPINK3调控肝细胞增殖的机理。首先用基因添加、干涉和抗体抑制等方法处理肝癌细胞、肝细胞及再生肝,然后用RT-PCR、Western Blot等方法检测上述材料的SPINK3及其受体EGFR和相应的信号通路相关基因和蛋白的变化,用生长曲线、流式细胞术等方法检测SPINK3对细胞增殖的影响,进而分析大鼠肝再生中SPINK3调控肝细胞增殖的机理,为了解正常细胞与肿瘤细胞、体内与体外的肝细胞增殖调控机理异同奠定基础。预期研究成果是发表SCI论文2-3篇。

英文摘要: Our laboratory discovered SPINK3 gene is closely related with hepatocyte proliferation when we studied the regulation of hepatocyte proliferation during rat liver regeneration. However, the mechanism of its regulation remains unclear. Therefore, this item will further study the effect of SPINK3 on hepatocyte proliferation. Firstly, we treated liver cancer cells, isolated primary hepatocytes and regenerating liver with the methods such as gene addition, gene interference and antibody inhibition, and then detected the expressive changes of genes and proteins of SPINK3 and EGFR signaling-related genes using the method such as RT-PCR, Western Blot and so on. Meanwhile, we detected the reffects of SPINK3 on cell proliferation with growth curve, flow cytometry, etc. This study set a basis for us to understand the similarities and differences of regulation mechanism of hepatocyte proliferation between normal cells and tumor cells, in vivo and in vitro. The results obtained in the course of the study are expected to publish 2-3 papers in SCI Journals.
