中文摘要: 二芳烯分子具有优异的光开关性能、热稳定性和耐疲劳性,在光电材料和生物医学成像领域具有广阔的应用前景。本课题利用全氟二芳烯优异的光开关性能,通过筛选闭环吸收波长在红外或近红外区域的二芳烯分子作为母体分子,并对其进行结构修饰,采用化学合成的方法在母体分子中引入与其闭环吸收波段范围相匹配的荧光发色团,进而构建荧光共振能量转移F?rster体,系统研究其在光辐射下的光物理和光化学变化行为,并将其作为荧光探针应用于生物荧光成像领域。利用目标分子在红外或近红外发光改善荧光探针的光穿透性;同时,利用其光辐射前后构型改变以及分子内能量共振转移导致的吸收光谱和荧光光谱显著变化的性能,将其作为荧光可逆探针,对染色的生物体荧光进行可逆调控,有效避免生物体内自发荧光的干扰,从而在荧光成像领域发挥重要的应用价值。本工作将拓展二芳烯研究的范围,为二芳烯材料的多功能化研究提供新的思路。

英文摘要: Diarylethenes have broad applications in photoelectric materials and medical imaging due to their superior optical switching property, thermal stability, and fatigue resistance. In order to further investigate perfluorinated diarylethenes as excellent optical switches, our research project will identify ideal perfluorinated diarylethenes, whose closed-ring isomer absorption is in infrared or near-infrared region, as parent molecules by structural variations and modifications. Fluorescent chromophores, whose emission range overlaps with absorption range of the closed-ring isomers of perfluorinated diarylethenes, will be linked to the parent molecules by chemical methods to construct fluorescence resonance energy transfer (FRET) F?rster pairs. Their photophysical and photochemical change behaviors upon UV radiation will be systemically investigated, and their applications in fluorescence bio-imaging will be explored. The new FRET pairs will improve light penetrating property of fluorescent probes in infrared or near infrared region. The changes of conformation, absorption, and fluorescence spectra of the FRET pairs before and after UV radiation, caused by the intramolecular resonance energy transfer, render the designed target molecules reversibility in modulation of fluorescent probes in cells.
