中文摘要: 本课题通过研究电针对骨性关节炎软骨细胞JAK-STAT信号通路表达的影响及电针后血清对TNF-α35825;导凋亡软骨细胞MAPK信号通路的影响,得出:(1)电针能有效增加骨性关节炎关节液中TGF-β#21547;量及促进关节软骨中STAT3、Smad3、LepRmRNA的表达,抑制关节液中COX2、iNOS、NO含量,延缓关节软骨的退变。(2)骨性关节炎经电针治疗后血清能有效抑制凋亡调控基因p38、FasmRNA,降低MAPK信号转导通路的表达,有效抑制TNF-α35825;导的软骨细胞凋亡。 临床电针治疗肾虚髓亏型膝骨性关节炎患者245例(279例膝),能有效缓解膝骨性关节炎患者的临床症状,抑制关节液中IL-1、IL-6、TNF-α12289;PGE2、MMP-3分泌,其中对Ⅲ#26399;膝骨性关节炎患者临床症状改善作用明显。 本研究发表学术论文6篇,其中SCI收录1篇;培养博士研究生1名,硕士研究生3名。

英文摘要: This Project studied on the effect of electroacupuncture on JAK-STAT signaling pathway expression of osteoarthritic chondrocyte and electroacupuncture serum on MAPK signaling pathways expression of TNF-αnduced chontroctye apoptosis, and concluded that Electro-acupuncture can effectively promote TGF-βbiological activity and inhibit COX2,iNOS and NO biological activity in synovial fluid, delay the morphology of articular cartilage degeneration; electroacupuncture serum can effectively inhibit apoptosis genes p38, Fas mRNA expression, reduce the expression of MAPK signal pathway, and inhibit TNF-αnduced chontroctye apoptosis. The clinical efficacy of electroacupuncture on treating 245 patients(279 knees) of knee osteoarthritis (KOA) of Shen-Sui insufficiency syndrome type found that electroacupuncture can effectively relieve knee osteoarthritis in patients with clinical symptoms, inhibit synovial fluid IL-1, IL-6, TNF-αPGE2, MMP-3 secretion, which is more effective ways to treat of knee osteoarthritis of stage Ⅲ This Project published 6 papers including SCI 1 paper, and trained 1 doctor graduate student and 3 master graduate students.
