中文摘要: 目前,有许多学者认为不完全表观重编程是体细胞克隆过程中克隆效率低下的原因。本研究以本实验室获得的一批转基因体细胞克隆绵羊为材料,拟通过对比雌雄2种细胞系来源的出生死亡和存活的转基因绵羊与正常同龄自然交配生产绵羊的各个组织(心脏、肺脏、肝脏、肾脏和肌肉组织)中8个印记相关基因(Peg10、Peg3、Dlk1、Gtl2、Cdkn1c、H19、Igf2r和Xist)的DNA甲基化状态,各选择3个个体进行研究,探究死亡和存活的转基因个体是否存在印记相关基因异常DNA甲基化修饰,推测死亡个体是否存在印记表观重编程不完全,进而寻找死亡绵羊死因;同时通过与本实验室已研究过的这8个印记基因在体细胞克隆绵羊个体中的DNA甲基化状态进行对比分析,推测外源基因的导入是否对8个印记相关基因DNA甲基化状态有影响,进而推测外源基因的导入对印记基因重编程的影响。通过本项研究进而为体细胞克隆及转基因体细胞克隆技术的应用

英文摘要: Currently, there are many scholars believe that the incomplete reprogramming the reason of low efficiency of cloning in the process of somatic cell cloning. In this study, our laboratory obtained a number of transgenic somatic cell cloning sheeps for the material. We proposed by contrast the DNA methylation status of 8 imprinted related genes(Peg10、Peg3、Dlk1、Gtl2、Cdkn1c、H19、Igf2r和Xist) in death and survival of transgenic sheeps, which derived from male and female cell lines, and normal bovine organizations (Heart, lung, liver, kidney and muscle tissue). Each group selectted three individuals to explore whether there is abnormal DNA methylation of imprinted related genes in death and survival of transgenic individuals, to suggest that the death of individuals whether related to incomplete imprinted epigenetic reprogramming, and then look for the death cause of dead sheeps. The results by contrast with the DNA methylation status of imprinted related genes in somatic cell clone sheeps, to speculate whether the exogenous gene affect imprinted genes DNA methylation status and reprogramming.
