中文摘要: 肿瘤标志物的高灵敏分析检测是提高恶性肿瘤早期诊断和治疗的关键。研究证实肿瘤患者外周血循环miRNA具有和恶性肿瘤相关的分子生物学特征,可作为敏感高效的肿瘤分子标志物,有望发展肿瘤无创诊断的新方法。该项目主要进行量子点标记探针用于外周循环血中标志物miRNA分子准确、灵敏、定量靶向诊断的应用基础研究,拟利用水相中合成的多色CdTe/ZnS核壳量子点偶联不同DNA分子作为荧光偏振信号探针,并结合指数放大系统及滚环扩增实现血液及细胞中的不同miRNA的同时测定,建立量子点标记探针对生物样品中miRNA分子靶向诊断的荧光偏振新方法,开发灵敏度更高、更方便快捷、可同时检测多种miRNA分子靶的血清学诊断新技术,提高肿瘤早期诊断的准确性,并研究生物样品中miRNA标志物准确量化结果与肿瘤发展之间的关系及对临床诊疗的潜在指导意义,为肿瘤的无损早期诊断及预后跟踪治疗提供可靠的实验支持与科学理论依据。

英文摘要: The studies on the analysis of cancer markers are the key to improve the early diagnosis and treatment of malignant tumor. More and more studies confirmed that RNA molecules of peripheral circulating blood from cancer patients have biological characteristics associated with malignant tumor, and they can be used as sensitive and efficient tumor molecular markers. Then miRNA is a potential marker to develop the noninvasive diagnostics of malignant tumor. In this project, combining multicolor of quantum dots with hybrid cycle amplification and rolling cycle amplification reaction we will develop a new fluorescence polarization sensor platform for simple, sensitive and multiplex analysis of miRNA. The method we proposed will provides a reliable experimental support and theoretical basis for the non-invasive early diagnosis and prognosis of tumor therapy, at the same time, it has important practical significance to illustrate the miRNA biological function.
