中文摘要: 咖啡作为世界三大饮料之首,有着重要的生产和科研价值。单核苷酸多态性是由单个碱基变异引起的DNA序列多态性,是第三代遗传标记物。开发利用单核苷酸多态性分子标记,可以对咖啡的种质资源进行多态性评价,可以探讨咖啡种属间的分类进化关系,进行分子标记辅助育种等。因此,单核苷酸多态性标记对于咖啡的遗传与育种具有重要的理论和实际应用价值。. 本研究拟以我国现有咖啡种质资源为材料,利用PCR-SSCP方法筛选分析SNP分子标记,对筛选出多态性引物的产物进行克隆测序,找出SNP位点,通过PCR-SSCP的谱带分型及测序结果进行咖啡种质资源的遗传多样性分析。

英文摘要: Coffee, is one of the three most popular beverages in the world, have important values in food production and scientific researches. SNP, as the third generation of genetic markers, are DNA sequence variations caused by changes in single nucleotide. Development and utilization of SNP markers could help evaluate the polymorphism levels of coffee germplasm resources, study the phylogenetic and evolutionary relationships, conduct marker-assisted selections. Therefore, SNP are very valuable theoretically and practically in coffee genetic and breeding researches.. In this research, coffee germplasm resources are used for the identification of SNP markers with the method of PCR-SSCP. The PCR products of the polymorphic primers are clone and sequence, yielding the SNP locus. Evaluate genetic diversity of the coffee germplasm resources are perform on the results from genotyping PCR-SSCP bands and sequencing of PCR product.
