中文摘要: 植物修复是一种高效、经济和生态友好的重金属修复技术。香根草作为世界知名的水土保持植物,因其综合抗性强、耐受并积累多种重金属等特点,在植物修复中的应用日益受到重视。为了科学评价香根草用于植物修复的潜能,揭示其重金属积累和耐受机制,本研究将探讨不同生态型香根草对重金属的耐性及其区隔化的特征。项目采用Weibull模型、结合重金属在根叶中的积累和光合速率、叶绿素荧光等光合生理生态指标,比较不同生态型香根草对重金属的耐性及积累特点,筛选用于植物修复的优质生态型。通过对重金属耐受型和敏感型香根草亚细胞组分中重金属含量的测定及原生质体中重金属分布的荧光观察,从细胞水平分析重金属在香根草细胞中的区隔化;并通过对参与重金属区隔化的2类重要基因HMA3、ABCC的克隆和响应分析,揭示重金属区隔化的分子生物学机制。

英文摘要: Phytoremediation is an effective, cheap and eco-friendly technology for heavy metal remediation. Vetiver grass (Vetiveria zizanioides (L.) Nash) can be used not for soil and water conservation, but potential also for phytoremediation, owing to unique morphological, physiological and ecological characteristics such as its tolerance to many kinds of extreme environment and high accumulation of heavy metal. In order to evaluate the phytoremediation potential of Vetiver grass, and to better understand its heavy metal accumulation and tolerance,differences in heavy metal tolerance and compartmentation between kinds of ecotypes Vetiver grass are investigated. The characteristic of heavy metal tolerance and phytoremediation potential of a range of Vetiver grass ecotypes are determined using Weibull model,heavy metal concentration of root and leaf, and photosynthetic physioecological characteristics.furthermore,Subcellular distribution of heavy metal is investigated, furthermore, the heavy metal distribution in protoplast is observed by fluorescent dye. and differences of compartmentation are compared in heavy metal tolerance and sensitive Vetiver grass ecotypes by above 2 experiments.HMA3 and ABCC are important vacuolar transporters for vacuolar sequestration of heavy metal compartmentation in plant cell.
