中文摘要: 树状大分子具有均一的三维球形纳米结构,尺寸可控,拥有内部空腔和大量的表面官能团,因而被广泛地用于药物输送。目前发展的树枝状大分子载体大多是基于聚酰胺-胺、聚丙烯亚胺、聚赖氨酸、聚酯等及其修饰物。这些树状大分子本身或其降解产物无治疗作用,因而只能起载体作用,完成药物输送任务后即成为病人的负担,必须排出体外。以多官能团的抗癌药物分子通过酯键来构筑可降解树状大分子,可获得兼具治疗和载体双功能的新型药物载体。本项目将以具有以抗肿瘤活性的没食子酸为基元,通过酯键构筑树枝状大分子载体。该载体在将药物输送入肿瘤细胞后将降解为大量没食子酸分子,继续作用于病灶,或与所载药物起到协同作用,强化所载药物的疗效。本课题将系统研究该树枝状大分子合成、载体结构与载药性能之间的关系,降解条件、降解速率与该双功能载体抗肿瘤能力之间的关系,为新型药物载体的设计与研究提供理论基础,并为设计新的药物载体提供新理念。

英文摘要: Dendrimers have been widely used in drug delivery because of their highly branched three-dimensional architecture, perfect monodispersity, nano sizes, high density of functional terminal groups, and controllable molecular weights. At present, dendrimer carriers are mostly based on poly(amidoamine), poly(propylene imine), polylysine, polyester and their derivatives. These dendrimers or their degradation products had no therapeutic effect and must be excreted which consumed additional physical of patients after drugs senting to tumor. Synthesis of biodegradable dendrimers with natural anticancer medicine itself will obtain drug carriers with both drug delivery and tumortherapy functions. This project will choose gallic acid which possesses anti-tumor activity as based unit to build a novel dendrimer carrier by ester bonds. After the drugs deliver into tumor site, the dendrimer carrier will degrade into large number of gallic acid molecules and continue to treat tumor therefore produce a synergistic effect with the drugs delivered by it. Systematic research on dendrimer synthesis, the relationship between structure and performance, carrier degradation conditions, degradation rate and anti-tumor capacity will be carried out.
