中文摘要: 炎症,损伤是冠心病研究领域的热点,白介素-8(IL-8)是参与心肌梗死的重要炎症因子,它可由炎症区域直接分泌,并进一步诱导产生,可贯穿于心梗前期、心梗急性期、再灌注损伤期、后期等,心梗再灌注时IL-8水平可反映再灌注损伤的程度。 课题组前期曾成功制备携IL-8单克隆抗体的靶向性声学造影剂,并成功用于粥样斑块的炎症反应,开辟了以炎症因子为介导的靶向性超声造影剂的新天地,在国内外为独家报道。此外课题组已将该造影技术成功用于兔心梗前期的观察。 本研究拟将携IL-8单克隆抗体的靶向性声学造影剂用于心肌梗死再灌注的兔模型,观察再灌注损伤的靶向性心肌造影特征。并使用超声波照射致微泡破裂,释放IL-8单克隆抗体,使其与再灌区域的IL-8中和,达到减轻炎症及相关连锁反应的目的。通过比较不同功率超声波照射的结果,探讨减轻心肌梗死再灌注损伤,控制心梗进程的方法,为早期干预治疗提供理论依据。

英文摘要: Inflammation and injury has become a hotspot in the field of coronary heart disease recently, interleukin 8 (IL - 8) is an important inflammatory factors involved in myocardial infarction, it can be directly secreted by the area of inflammation, and further induced other inflammatory cytokines, it also throughout the early myocardial infarction, acute myocardial infarction and reperfusion period and later period .The levels of the IL - 8 can reflect the degree of reperfusion injury during reperfusion period. We have prepared targeting acoustic contrast agent with IL - 8 monoclonal antibody, and used it in atheromatous plaque successfully. It opened up a new world of mediated inflammatory factor for targeted ultrasound contrast agent, and it is the scoop both at home and abroad. Moreover, this imaging technique has been successfully used for observation of early myocardial infarction in rabbits. IL - 8 monoclonal antibody targeting acoustic contrast agent will be used in rabbit model of myocardial infarction, then we will observe the imaging characteristics of myocardial reperfusion injury.At the same time, this research proposed by ultrasound-targeted microbubble destruction and the release of IL - 8 monoclonal antibody, to promote IL - 8 monoclonal antibody combining with IL - 8, By comparing the results of the different power ultrasound irradiation, to find out the way of reducing myocardial infarction reperfusion injury, and ways to control the process of myocardial infarction, as to provide theoretical basis for early intervention treatment.
