中文摘要: 针对AD进行性神经改变的复杂性、不确定性等特点,围绕AD临床诊治中遇到的问题,运用信息论、复杂网络理论、计算实验等理论和方法,对脑功能网络建模与分析方法、AD进行性神经改变机制、以及AD早期诊断及神经功能评价等问题进行研究:根据实验范式设计,采集AD不同阶段静息态功能磁共振影像;研究新的基于信息论的脑功能网络建模与分析方法,以建立高重测信度、更加逼近大脑实际工作机制的脑功能网络,探索新的更符合神经生理学的网络模块化算法及Hub节点定义;建立基于计算实验的AD脑功能网络演化模型,以模拟脑网络层面AD进行性神经改变的动态演化过程,获得对AD神经病变进程更完整、更全面的认识和理解;基于上述研究,构建基于大脑网络尺度的AD早期诊断及神经功能评价的量化模型,为AD辅助诊断与临床治疗提供新的方法和技术支持,具有潜在的临床应用前景。本项目的研究方法和思路可以推广到其它神经疾病的研究,具有广泛的研究意义。

英文摘要: Aiming at the complexity and uncertainty progressive neurological changes after AD and the issues encountered in the diagnosis and treatment of AD,this project makes use of information theory, complex network theory and computational experiments method to investigate modeling approaches and analytical methods of functional brain network, the mechanism of progressive neurological changes after AD,the early diagnosis of AD, and the evaluation of neurological function after AD. According to our experimental paradigm designed, the resting-state fMRI data derived from each period of AD will be acquired. Based on information theory, we explore new modeling and analysis methods of functional brain network which are applied to establish functional brain network with high test-retest reliability and better approximation to the actual mechanism of brain network. Afterwards, we will explore novel algorithm of network modularity and new definition of Hub nodes which is more in line with neurophysiology. On the basis of computational experiments, functional brain network evolution model will be built in order to simulate the dynamic progressive neurological process after AD at the brain network level. Then, we will get more comprehensive knowledge and understanding on the AD.Based on the investigations above, a quantitative model will be built for early diagnosis and neurological function evaluation of AD according to the scale of brain network.Our study, providing new methods and technical support for AD diagnosis and clinical treatment, have potential prospects for clinical applications prospect.It is of significance to extend the research methods and ideas in this project for the study of other neurological diseases.
