中文摘要: 炎症信号传导网络在炎症反应中起着至关重要的作用。理解炎症网络应对刺激信号的动力学行为及其分子机制,对预防和治疗慢性炎性疾病及相关癌症具有重要意义。 炎症性质(急性-慢性)的变化以及恶性转化涉及从微观(分子)、介观(细胞)到组织、器官(宏观)多种层次,在本质上是多尺度的。因此,其调控动力学极其复杂。尽管临床和生物化学实验研究已取得巨大进展,然而炎症网络应对刺激信号的动力学性质变化的分子机制仍然缺乏系统了解。为此,本项目提出一种整合的多尺度建模和计算模拟方法,研究不同层次上炎症演变及恶性转化的动力学行为,并把宏观和介观层次上可以观察的性质和动力学变化与微观层次上分子之间的相互作用有机地联系起来,从而有可能达到理解其分子机制的目的。本项目的研究可能为发现预防和治疗慢性炎症及其有关癌症新方法提供理论基础。

英文摘要: Inflammatory signal transduction networks play a vital role in the inflammatory response. It is of great importance to understand the dynamic behaviors of the inflammatory networks and their molecular mechanisms for the prevention and treatment of chronic inflammatory diseases and cancer.The state transition (from acute to chronic) of inflammation and its malignant transformation occur at multiple levels ranging from microscopic (molecules), mesoscopic (cells) to macroscopic (tissues and organs). The dynamics of inflammation-cancer is thereby multi-scale in nature and is highly complex as well. Although significant progress has been made in clinical and biochemical experiments, the systematic understanding of the molecular mechanisms is still lacking. This project suggests an integrated multi-scale modeling and computer simulation method to study the state transition of inflammation and its malignant transformation, and to link the properties and changes of inflammation and cancer, which can be measureed experimentally at the macroscopic and mesoscopic levels, to molecular interactions at the microscopic level. As such, it provides a possible route to understand molecular mechanisms of the state transition and malignant transformation of inflammation.
