中文摘要: 本项目提出了一种新颖的能够直接取样的微纳毛细管色谱-电化学检测系统,具有锥形尖端的微纳毛细管可实现双重功能:在电渗泵驱动下实现飞升到皮升级别的定量进样、在压力流驱动下实现分离。微纳毛细管的末端制备有电极,所分离的物质在毛细管末端进行电化学检测。此分离检测系统可以实现对微小体积的样品直接进样,尤其是直接进行活体单细胞的分析,避免了复杂的进样管道和进样阀的控制。在时间和空间上动态获得细胞内的神经递质的变化、药物代谢等,对于生物学和医学领域的研究具有重要的意义。

英文摘要: We propose to develop a novel micro/nano-capillary chromatography-electrochemical detection system, which can achieve directly sampling injection. The micro/nano-capillary with a pipet-tip on the end can realize dual functions: a valve for fL/pL sample injection via an electroosmotic pump and a separation column under a pressure-driven flow. A working electrode is prepared at the other end of the micro/nano-capillary for detection of the separated analytes. This system eliminates external auxiliary tubes and reservoirs for sample injection, can be used to directly inject small volume of samples, especially to achieve living single cell analysis. This system can be used to study the chemical information in cellular fluid, the changes of neurotransmitters, drug metabolism, etc., which has great significance for biological and medical research.
