中文摘要: 组蛋白分子伴侣结合组蛋白,介导在核小体组装/去组装过程中组蛋白与DNA的相互作用,是染色质表观遗传调控的重要机制之一。不同类别的分子伴侣对于组蛋白底物有着不同的偏爱性。通过遗传、细胞、分子生物学等方法和手段,我们从细胞周期调控、DNA损伤及修复、细胞分化几个方面对植物主要的几类组蛋白分子伴侣,即CAF-1、ASF1、NRP和NAP1,展开了功能分析。通过分析相应组蛋白分子伴侣缺失突变体在植物根器官发育过程中的缺陷,研究组蛋白分子伴侣作为染色质相关因子,参与调控植物根细胞增殖和细胞分化的分子机理,以及不同组蛋白分子伴侣相互之间功能的互作关系。

英文摘要: Histone chaperones bind histones, mediating interaction of histones with DNA during nucleosome assembly/disassembly, and function as one of the important mechanisms of epigenetic modulation on chromatin. Different histone chaperones have different preference to their histone substrates. We carried out functional characterization of several important histone chaperones, CAF-1,ASF1,NRP and NAP1, in the aspects of cell cycle regulation, DNA damage/repair and cell differentiation by genetical, cytological and molecular means. By analyzing the growth defects of plants mutant in histone chaperones, we studied the molecular mechanism by which the histone chapeornes, as chromatin-related factors, participate in the proliferation and differentiation of plant root cells, as well as the functional intersection among these chaperones.
