中文摘要: 有机磷杀虫剂的毒性之一是对生物体产生氧化胁迫,SOD在抵御氧化胁迫中发挥着重要作用。本项目以中华稻蝗为对象,研究有机磷杀虫剂对转录因子的影响,进而阐明其对SOD基因的转录调控机理。主要研究内容有:1)在RACE法获得SOD基因全长序列的基础上,进行RNA干扰研究,结合马拉硫磷和毒死蜱染毒,测定阴离子自由基含量,筛选出起重要作用的SOD基因。2)通过Genome-walking方法克隆其启动子区序列,并分析其特性。3)通过SOD基因启动子元件缺失、构建启动子-报告基因瞬时表达载体、转染昆虫细胞,用马拉硫磷和毒死蜱处理昆虫细胞,测定荧光素酶活性,确定各个启动子元件在SOD基因转录调控中的作用;将核蛋白抽提液与标记的转录因子结合区多聚核苷酸序列进行孵育,通过凝胶电泳迁移率方法,确定转录因子与SOD基因启动子区的结合特性,阐明有机磷杀虫剂对SOD基因的转录调控机理,为中华稻蝗的有效治理提供依据。

英文摘要: Oxidative stress is one of the toxicity of organophosphate insecticides. SOD plays important roles in defense against oxidative stress. This project will study the effects of organophosphate insecticides on transcription factors and illustrate the mechanism of transcriptional regulation of SOD in Oxya chinensis. First, important SOD genes will be selected using RNA interference technique based on the sequence of full-length of SOD genes (RACE Amplification Kit). Second, promoter sequences of the selected SOD genes will be cloned using genome-walking technique and the characteristics will be analyzed using relative softwares. Then, the roles of promoter elements of SOD genes will be determined by generating a series of 5' deletions, constructing luciferase reporter plasmids, transfecting insect cells, treating cells using malathion and chlorpyrifos, and measuring the luciferase activities. Meanwhile, the mobility shift assay will be carried out with the complexes of double-stranded oligonucleotied probes end-labeled with [γ-32P]ATP and nuclear extracts from cells. By these means, we will illustrate the mechanism of transcriptional regulation of SOD gene by organophosphate insecticides. This work will provide a reasonable basis for the effective management of Oxya chinensis.
