中文摘要: 生物单分子研究是以生命科学问题为核心的、多角度、多学科交叉的新兴研究领域,需要发展适合单分子水平研究的科学理论和技术。本项目应用基于原子力显微镜的纳米操纵技术,结合单分子PCR技术,探索了在固相表面和液体环境中单个DNA分子连接的可能性。初步建立了固相表面上单个DNA分子连接反应的方法,并在实验的基础上,对影响单分子反应的多种因素进行了细致的分析,探索了界面上DNA生化反应机理、界面对DNA性质及其行为的影响、以及低拷贝DNA扩增的有效性,为解决DNA"分子手术"中的关键技术问题提供了多方位的技术支持。

英文摘要: Biological single-molecule study is an emerging multidisplinary research area based on the life sciences. Its unique property need us to develop novel scientific theories and technologies suitable for it. This project, combinding the nano-manipulation technique base on atomic force microscope with single-molecule PCR technique, was used to investigate the feasibility of ligating single DNA molecules on surfaces and/or in liquid environments. The single-molecule ligation reaction on surfaces was proposed and explored. Based on experimental data, detailed analysis of a variety of factors affectng single-molecule reactions on surfaces was perfomed. It will be useful to understand the mechanism of DNA biochemical reactions, properties and behavior of DNA on interfaces.Meanwhile, the efficiency of low-copy DNA amplification was also explored. These technologies will be helpful to solve the key problems of "DNA molecular surgery".
