中文摘要: 本项目面向我国重大疾病预警与诊断新方法与新技术的战略需求,针对化学生物传感领域国际前沿课题,以高灵敏、高选择性卵巢癌循环肿瘤细胞电化学生物传感器的构建及临床诊断与预后判断应用为目标,通过学科交叉(包括分析化学、电子学、界面化学、分子生物学及临床医学等),开展原始创新的综合集成研究。项目的主要研究内容为:结合核酸适体这一新型的生物识别单元及其生物识别事件所独特的生物化学性质,发展基于高灵敏场效应传感技术、超分子自组装电化学通道技术、以及基于毛细管电化学微通道技术的细胞检测信号转换放大新机制,构建基于循环肿瘤细胞-核酸适体特异性识别的恶性肿瘤,如卵巢癌的快速、可靠、低成本、高通量的早期诊断生物传感新技术与器件。这些研究成果对于卵巢癌等恶性肿瘤的早期预警和临床诊断具有重要的意义,研究成果有广泛的理论价值与应用前景。

英文摘要: In the present project, surrounding the strategic requirements for new methods and techniques in diagnosis and early alert of grave diseases, and aiming at the international frontier of chemo/biosensing, the pioneer fundamental research will be carried out. This research will focus on fabrication of electrochemical biosensor to detect ovarian cancer circulating tumor cells with high sensitivity and selectivity and apply to clinical diagnosis and prognosis. Through the knowledge conjugation such as analytical chemistry, electronics, interfacial chemistry, molecular biology and clinical medicine, etc., we carry out the integrated study of original innovation. The main contents of this project is : 1) Combining with the aptamer, a novel bio-recognition element and its unique biochemical property in biorecognition, via high sensitive field effect sensing technology, electrochemical channel of supramolecular self-assembly technology and electrochemical microchannel of capillary technology, we develop novel signal amplification technology to detect cells. Based on the specific recognition between circulating tumor cells and aptamer, we develop biological sensing technology and devices with the characteristic of super-rapid, high selectivity, low-cost and high throughput for the early diagnosis of tumor such as ovarian cancer in cell level. These research progeny will be very important for the early alert and clinical diagnosis of ovarian cancer and other tumors. Moreover, the research progeny have wild application and important theoretic value.
