中文摘要: 动物源食品中抗生素残留问题关系到食品安全和出口贸易,对食品安全控制和检测技术提出很高的要求,而现有方法存在很大局限性。本项目拟将生物传感分析、纳米器件构建、快速检测技术及适配体优选技术联合,首先通过对已报道RNA适配体改造和ssDNA适配体筛选结合的方法获得适合抗生素规模化检测应用的适配体文库;其次在检测策略设计中以适配体为主,辅以抗体和特定酶作为识别元件,以纳米材料为载体构建识别-标记-放大和表征体系,以电化学和光谱学方法为主要手段,通过不同标记物对靶分子有效标记,对检测信号进行有效放大和表征,发展出多种高灵敏度、高选择性、低检测限的方法;最后通过考察方法的稳定性和重复性,并通过对功能化丝网印刷电极和干试剂技术、整合型便携式电化学检测技术和基于功能化AuNPs或适配体复合物的试剂盒型检测技术等的运用,建立起对食品抗生素残留规模化快速检测的平台,实现对实际样品的高效检测。

英文摘要: Antibiotic residues in animal derived food is not only concerned with food safety, but also export trade. Therefore, methodologies and techniques of food safety control and residues detection need to be highly developed, due to the limitations and shortcomings in current methods. In this project, techniques in bioassay, fabrication of nano-devices, fast detection and aptamer screening will be applied in a combined format. Firstly, aptamer library for scalable antibiotics detection will be founded through modification of reported RNA aptamer or screening ssDNA via SELEX procedure. Then a variety of detection strategies will be designed, where aptamers together with some antibodies and special enzymes are used as recogniation elements, different nanomaterials are used as carriers of recognition-marker-amplification detection systems. Based on the effective mark manner by different probes and signal amplification mechanisms, detection methods with high sensitivity, high selectivity, and low detection limit can be developed by using electrochemical and spectral technologies. Finally, after studying on the stability and repeatablity, scalable and rapid detection systems for antibiotic residues in food can be established by using screen-printed electrodes and dry reagents techniques, portable electrochemical devices,
