中文摘要: 肿瘤间质中浸润的巨噬细胞(TAM)在非小细胞肺癌(NSCLC)侵袭转移中的作用尚存争议。不同研究的结果并不一致,甚至相反。尽管有报道提示TAM存在抑制和促进肿瘤发展两大亚群,然而如何鉴别这些亚群至今尚无有效的标记物。本课题组前期研究肿瘤细胞骨桥蛋白(OPN)与NSCLC转移关系时,发现约45%患者肿瘤间质中TAM也表达OPN,进一步分析显示这部分患者较OPN阴性TAM患者更容易出现远处转移,预后较差。OPN阳性TAM在NSCLC的生物学功能目前尚无报道。据此,我们推测OPN可能是促肿瘤TAM的生物标记物之一。为此,我们拟借助体外细胞共培养模型、OPN基因敲除裸鼠骨髓细胞移植模型、荧光标记骨髓移植模型等研究手段,探讨OPN对NSCLC间质TAM生物学功能的调控。

英文摘要: The role of tumour-associated macrophages (TAMs) in NSCLC is not clear because the data for NSCLC are contradictory. Although it has been reported that TAMs include two clones (M1 and M2) which inhibit and promote tumor progress respectively, there are no exact biomarkers to identify these clones effectively. OPN positive infiltrating TAMs were detected in 45% of patients with NSCLC when we studied the relationship between OPN and tumor metastasis in NSCLC. We found that patients with OPN positive infiltrating TAMs in tumor tissues were associated with distant metastasis and poor prognosis. However, there is no report to elucidate the biological functions of OPN positive infiltrating TAMs in NSCLC. So we hypothesize that OPN is a biomarker of TAMs wich promote tumor progress.In this study,we investigate the regulation of OPN in TAMs of NSCLC by a co-culture model in vitro, bone marrow transplantation model in OPN konckout nude mice, fluorescence labeling of bone marrow transplantation mode.
